Funny you should ask this.
I just had the second puncture of my life just the other day (tiny tiny thorn), in the same tire as the first. The first one was brutal, a screw, but both times the tube was patched, the offending article was removed, any debris was cleaned out and the tube replaced. The fact that this tire had been previously punctured by the screw didn't make it any more vulnerable in general, according to my repair guy ( @StevenKennard@zeroes.ca ). The thorn found a new spot.
Two punctures in a month in 71 bike riding years. Can't complain, I guess.
