My thinking is: 1: the patch and glue will be curing in the curved, expanded shape where they'll be in use, so if there is any flexibility issues, they'll be preemptively addressed. 2: the inflation presses the patch to the tube nice and evenly 3: if there's a problem with the patch, or glue, or another puncture you didn't notice, you know because the tube didn't hold pressure overnight. Better to find this out before putting the patch to the test on a real ride.
